Radial immunodiffusion or the Mancini method is a technique used to demonstrate or measure the amount or concentration of an antigen in a culture. The antibody is added to liquid agar and this is poured onto a petri dish and forms a gel. Then the antigen is added in different concentrations in small Wells. A circle will now form from the Wells whose diameter is a measure of the reaction.
The kit contains: antigens and antibodies, petri dishes, well formers, agarose, buffer and microreagent tubes.
Also needed: micropipettes, water bath, microwave or hot plate, incubator, pipettes, rulers and distilled water.
